Antidiabetic Activity Assay Service

Some marine glycoproteins and glycans have antidiabetic activity, such as algal glycoproteins and seaweed polysaccharides. The determination of the antidiabetic activity of marine glycoproteins and glycans is a unique examination process. This process uses different strategies to evaluate the impact of marine-derived glycoproteins and glycans on biochemical aspects associated with diabetes. CD BioGlyco provides in vivo and in vitro antidiabetic activity assay services, aiming to comprehensively evaluate the impact of marine glycoproteins and marine glycans on diabetes. In vivo experiments observe the overall physiological response of animal models, while in vitro experiments use cell models to deeply explore the mechanism of action. In vitro antidiabetic activity assay services we provide:

  • Establishment of cell models: Use diabetes-related cell models, such as pancreatic beta cells, adipocytes, liver cells, etc., to simulate the pathophysiological state of diabetes.
  • Treatment of marine glycoproteins/marine glycans: After co-culturing marine glycoproteins or glycans with cell models, observe their effects on cell functions.
  • Determination of glucose metabolism-related indicators: Use specific experimental methods, such as a glucose-stimulated insulin secretion test, to evaluate the impact of marine glycoproteins/glycans on cellular glucose metabolism.
  • Determination of enzyme inhibition: Evaluate the inhibitory effect of marine glycoproteins/glycans on related enzymes, such as α-amylase, α-glucosidase, pancreatic lipase, and dipeptidyl peptidase IV (DPP-IV).
  • Observation of cell function and morphology: Use a microscope to directly observe cell morphology to understand the impact of marine glycoproteins/glycans on cells.
  • Measurement of other indicators: Depending on the purpose of the research, other diabetes-related cellular indicators are also measured, such as insulin sensitivity, fatty acid uptake, etc.

In vivo antidiabetic activity assay services we provide:

  • Establishment of animal models: Select appropriate diabetic animal models, such as diabetic mice, rats, or non-human primates. These models are established through methods such as genetic engineering, chemical induction, or high-fat diet.
  • Administration: Marine glycoproteins or glycans are administered to animals at different doses and frequencies. Administration methods include oral administration, intraperitoneal injection, or other routes, which are determined based on experimental needs and the characteristics of the animal.
  • Monitoring of physiological indicators: Collect animal blood samples regularly to measure physiological indicators such as blood sugar, insulin, and glucagon. Then observe and record relevant data of animals in each group.
  • Tissue sampling and pathological examination: After the experiment, tissue samples are taken from the animals, such as pancreatic islets, liver, adipose tissue, etc. Pathological examination is performed to observe histomorphological changes.
  • Measurement of other indicators: Depending on the purpose of the research, other physiological indicators related to diabetes, such as blood lipids, inflammatory factors, etc., are also measured.

Results: The authors performed a screen for potential medicinal properties of crude phyllotannins derived from the perennial brown seaweed Eiseki bicyclica (CPEb). The study aimed to explore their effectiveness in the treatment of diabetes. It was found that CPEb enhanced the uptake of 2-NBDG in C2C12 myotubes in both standard conditions (16.08%) as well as when stimulated by insulin (51.09%). It was also shown to promote IRS/AKT-mediated glucose absorption, whilst simultaneously activating the AMPK pathway. The observed anti-a-glucosidase activity of CPEb underscores its potential application in diabetes therapy.
