Results: The authors described the structure and function of a marine sponge-derived thrombopoietin receptor (MPL) agonist called thrombocorticin (ThC). ThC was identified as a homodimerized lectin with calcium-dependent fucose-binding properties. Their detailed characterization of lectin-induced activation revealed that, like oncogenic activation, glycan chain-mediated activation continued due to restricted receptor internalization. The observed strong synergy between ThC and thrombopoietin indicated that ThC facilitated the formation of receptor dimers on the cell surface.
