Sample Preparation

  • Sample collection: We collect marine mangrove polysaccharide samples from reliable sources.
  • Separation and purification: We perform necessary purification and separation treatments on the collected samples to remove impurities and improve the purity of the samples. Then we use solvent extraction, enzyme digestion, and other state-of-the-art methods to effectively separate and purify the polysaccharides.

Structural Characterization

  • Infrared spectroscopy analysis (IR): We use infrared spectrometers to quickly and accurately identify the functional groups in marine mangrove polysaccharides, providing important information for the structural analysis of polysaccharides.
  • Nuclear magnetic resonance spectroscopy (NMR): We use one-dimensional and two-dimensional NMR techniques to accurately analyze the glycosidic bond type, monosaccharide composition, and connection order of polysaccharides, thereby constructing a molecular structure model of polysaccharides.
  • Mass spectrometry (MS): Combined with MS technology, we further verify the molecular weight and distribution of polysaccharides, as well as possible glycan chain modifications.

Characterization of Physical and Chemical Properties

  • Molecular weight determination: We use methods such as gel permeation chromatography (GPC) or laser light scattering (LLS) to accurately determine the molecular weight and distribution range of marine mangrove polysaccharides.
  • Thermal property analysis: We use techniques such as thermogravimetric analysis (TGA) and differential scanning calorimetry (DSC) to evaluate the thermal stability, melting point, and glass transition temperature of polysaccharides.

Bioactivity Evaluation

  • In vitro bioactivity test: We establish a series of in vitro bioactivity test systems, such as antioxidant, antibacterial, and antiviral experiments, to evaluate the bioactivity of marine mangrove polysaccharides.
  • Cell experiment: We use cell culture technology to study the effects of polysaccharides on biological processes such as cell proliferation, apoptosis, and differentiation, and further verify their bioactivity.

Publication

Technology: Extract and purify Rhizophora mucronata leaves polysaccharide, RP-HPLC

Journal: South African Journal of Botany

IF: 3.1

Published: 2022

Results: The authors extracted and purified Rhizophora mucronata leaves polysaccharide (RMLP) from mangrove R. mucronata leaves using anion-exchange and size-exclusion column chromatography. They evaluated the physicochemical characterization of RMLP with spectroscopic analysis and RP-HPLC. The molecular weight of RMLP was determined to be 2.69 kDa, consisting of monosaccharides such as arabinose, galacturonic acid, galactose, mannose, xylose, rhamnose, and glucose. Viscosity analysis indicated the polysaccharide exhibited pseudoplastic fluid behavior with shear-thinning properties.
