Relative Quantification Analysis of Marine Glycoprotein

Relative protein quantification is a mass spectrometry-based technique. The purpose of relative quantification is to compare the relative abundance of proteins in each sample. By resolving differences in protein expression in organisms under different conditions, the role of proteins in various biological processes is clarified. We have a diverse range of isotope labeling quantification techniques, including isotope-coded affinity tags (ICAT), isobaric tags for relative and absolute quantitation (iTRAQ), etc. Combining mass spectrometry with labeling techniques, relative quantitative analysis of glycoproteomics is achieved by enzymatic cleavage, isotope labeling, separation, and detection of proteins.

With the in-depth study of glycosylation modifications, mass spectrometry-based methods for glycosylation analysis are greatly developed. Non-labeled mass spectrometry quantitative analysis methods also achieve relative quantitative detection of glycoproteins. We also provide a relative quantitative analysis of marine glycoprotein based on matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS).

Absolute Quantification Analysis of Marine Glycoprotein

Absolute quantitative analysis is a targeted quantitative proteomics technique. This technique is used for absolute quantification of proteins and their modification states. We use multiple reaction monitoring (MRM) or selective reaction monitoring (SRM) to determine the absolute content.

Results: In this study, manatee serum proteomics was analyzed using iTRAQ liquid Chromatography with tandem mass spectrometry (LC-MS/MS) and 2D-DIGE. By this method differentially expressed proteins in the serum of manatees affected by different mortality events were identified. The results showed increased levels of several proteins in the serum of manatees from the Indian River Lagoon compared to controls, including histidine-rich glycoprotein, among others. This study demonstrates that glycoproteomics analysis can be used to analyze disease-related proteomes.

Applications
