We take the samples to be tested and culture them together with different and already activated strains, and at the same time, set up a blank control group. Then, the optical density value of the culture solution at around 600 nm, as well as the pH value, are detected using an ultraviolet-visible spectrophotometer. The probiotic activities of marine glycans and glycoproteins are analyzed based on the growth curves.

Different pH environments affect the probiotic activity of marine glycans and glycoproteins. Firstly, we test the low pH tolerance of marine glycans and glycoproteins. We use a hydrochloric acid solution to adjust the pH of the liquid medium to lower values for marine glycans & glycoproteins and specific bacteria, respectively. The glycans and glycoproteins to be tested are incubated with the specific bacteria, while a blank control group is set up. Finally, the optical density of each group is measured at 600 nm to analyze the low pH tolerance and bacterial inhibitory activity of the samples to be tested.
We add bile salts to the broth culture separately to get different concentrations of bile salt solutions. The samples (marine glycans and glycoproteins) are to be tested and specific bacteria are added to them separately, mixed, and incubated. Meanwhile, a blank control group is set up. Finally, the absorbance of each group is measured to analyze the bile salt tolerance of the samples to be tested.
Antibiotic susceptibility testing is an important element in the in vitro safety assessment of existing and potential marine glycans and proteins. We offer a variety of experimental methods to test substances for antibiotic susceptibility. Examples include antimicrobial gradient strips, agar dilution methods, tube-dish diffusion methods, and a variety of polymerase chain reaction methods. Secondly, we also offer the concentration gradient test strip method to determine the lowest inhibitory concentration of the sample to be tested to distinguish between natural and acquired resistance.

Results: In this research, the authors utilized a mouse model of dextrose sodium sulfate (DSS)-induced ulcerative colitis to characterize the beneficial properties of Enteromorpha clathrata polysaccharides (ECP). High-throughput sequencing and bioinformatics analyses showed that ECP significantly altered the structure of the intestinal microbiota and increased the abundance of Paramecium spp. Furthermore, the research found that Parabacteroides distasonis had an anti-ulcerative colitis effect on DSS-fed mice. In this research, the beneficial effect of ECP on ulcerative colitis was demonstrated for the first time.
