Raw material pretreatment

Washing and crushing: Our laboratory dusts and cleans the collected sea cucumbers. The dried sea cucumbers are processed into a powder suitable for extraction by mechanical crushing equipment.

Extraction and Isolation

  • Organic solvent extraction: We repeatedly extract the sea cucumber at least three times at room temperature using ethanol. Our researchers suspend the ethanol extract in water and then separate it with petroleum ether and n-BuOH, respectively.
  • Chromatography purification: Our common separation techniques include silica gel chromatography, reversed-phase abbr high-performance liquid chromatography (RP-HPLC), thin layer chromatography (TLC), and size exclusion chromatography (SEC). It is difficult to completely purify a single component with a single technique, so we offer a combination of techniques to achieve high-purity separations.

Characterization

  • Basic testing: Our lab provides professional purity testing by HPLC. We provide accurate determination of monosaccharide composition by gas chromatography-mass spectrometry (GC-MS).

  • Bioassay: We measure the antifungal activity of variegatusides using a variety of strains. The strains we provided include but are not limited to Candida albicans, Candida parapsilosis, Cryptococcus neoformans, Candida tropicalis, Candida pseudotropicalis, and Microsporum gypseum. Our researcher use an automatic microplate reader instrument in the determination of optical density (OD) to evaluate its antifungal activity.

Results: In this work, researchers extracted a total of seven glycosides from sea cucumber, including four novel non-sulfated triterpene glycosides variegatuside C-F and three known glycosides. The researchers characterized the structures of the glycosides by NMR and ESI-MS. In vitro activity tests showed that all of these trip glycosides have very significant antifungal activity. This study provides valuable information on antifungal natural products.
