Marine Glycosidase Analysis Service

Marine glycosidase, a subtype of hydrolytic enzymes, plays an instrumental role in catalyzing the hydrolysis of glycosidic bonds in complex sugars. Being prevalent in marine ecosystems, these remarkable enzymes are critical in decomposing organic material, contributing to the carbon cycle.

In addition, we use mass spectrometry-based technology to identify glycosidase, including protein sequence analysis and identification of post-translational modifications.

Sequence and Phylogenetic Analysis of Marine Glycosidase: We provide comprehensive sequence and phylogenetic analysis of marine glycosidase to understand the evolutionary relationships among organisms and to predict the properties of a new or altered gene. Results: The authors reported a new glycoside hydrolase family 2 (GH2), named AqGalA, isolated from a marine alga-associated Bacteroidetes organism, notable for its broad substrate specificity. The biochemical analysis showed that AqGalA efficiently broke down β-galacturonide, β-glucuronide, and β-galactopyranoside through retaining mechanisms. The structure of AqGalA in complex with galacturonic acid (GalA) was ascertained by crystallography, and subsequent mutagenesis helped determine that the charge traits at the uronate-binding subsites played a role in determining substrate selectivity for uronide hydrolysis. The authors proposed that the conformational flexibility of the AqGalA active-site pocket was a crucial factor for the broad substrate enzyme selectivity.
