Results: In this study, glycoproteins and glycopeptides in Phaeodactylum tricornutum were analyzed using LC-MS/MS. The researchers extracted the glycoproteins from the algal cells after grinding and lysing them. Then they analyzed and classified the N-glycopeptide sequences through digestion and N-glycopeptide enrichment. A total of 863 different N-glycosylated peptides were eventually identified. Most of the identified N-glycopeptides were in the range of 9-23 amino acids in length. This study provides a method for the analysis of N-glycopeptides and the exploration of N-glycosylation pathways in P. tricornutum.
