Animal Material Collection

In our lab, we have professional specimen collectors to help clients collect animal samples from all over the world. We provide sample collection services according to the needs of our clients. Scabraside A and scabraside B are isolated from the sea cucumber Holothuria scabra (Holothuriidae) collected from the South China Sea. Scabraside D is extracted from the sea cucumber H. scabra collected from Andaman Sea, Krabi Province, Southern Thailand.

Extraction and Isolation

After grinding the collected tissues into powder, our researchers obtain the crude extract by organic solvent method and resin column. Combined with silica gel columns and high-performance liquid chromatography (HPLC), our lab provides high-purity scabraside.

Identification and Analysis

Based on a reliable structural analysis team, we provide monosaccharide composition analysis, acid hydrolysis, gas chromatography-mass spectrometry (GC-MS) analysis, nuclear magnetic resonance (NMR) spectroscopy analysis, infrared spectroscopy (IR) analysis, MS/MS analysis.

Bioassay

We have a wide range of cell and animal models to meet our clients' needs in scabraside activity testing. For example, we have human leukemia cells, human lung cancer cells, and human hepatoma cells.

Results: In this study, researchers found that triterpene glycosides in sea cucumber extracts have strong anticancer properties. Combining database searches and molecular interactions analysis, the researchers evaluated the antilung cancer properties of intercedenside C, scabraside A, and scabraside B under in silico conditions. Ultimately, all three compounds showed superior anticancer properties. This also provides potential targets for the development of novel anticancer drugs.

What sugars do scabraside A contain?

The sugar moiety of scabraside A includes D-xylose (Xyl), D-quinovose (Qui), D-glucose (Glc), and 3-O-methylglucose (MeGlc).

Why combine multiple techniques to isolate saponin monomers?

There are a series of saponin components with similar structures and subtle differences in the same species, such as scabraside A, B, and D. The crude extract of sea cucumber contains many impurities, even though only one spot on the thin-layer chromatographic plate. After instrumental analysis, it may be composed of multiple compounds. The purification effect of a single chromatographic technique is not satisfactory, and several combined chromatographic techniques are needed to achieve a better purification effect.
