Results: The authors aimed to characterize and evaluate the anticoagulant potential of sulfated polysaccharides (SP) extracted from the green algae Halimeda opuntia. Four SP-rich fractions: F23, F44, F60, and F75 were obtained through proteolytic digestion using papain, followed by ethanol precipitation. Agarose gel electrophoresis confirmed the presence of SP, revealing different populations within each fraction. Among these, the F44 fraction stood out, yielding 5% of the initial algae weight and demonstrating anticoagulant activity via the activated partial thromboplastin time assay, which measures the intrinsic/common coagulation pathway. Interestingly, further purification of F44 resulted in prothrombin time activity, alongside the enhanced activated partial thromboplastin time activity. The anticoagulant activity of P1F44 was found to be dependent on coagulation factors IIa and Xa, with a higher potency observed via factor IIa.
