Sulfated Fucan-based Mimetic Development Service

Sulfated Fucan-based Mimetic Development Service

CD BioGlyco has professional Marine Carbohydrate Mimetic design, synthesis, and analyst groups, developers all have rich industry experience, equipped with advanced instrumentation facilities to provide clients with a one-stop sulfated fucan-based mimetic development solution strategy. Sulfated fucan-based mimetic consists of α-1,3 or alternating α-1,3/1,4-linked D-L-fucose backbone. Fucoidan from Sargassum thumbergii, Fucus vesiculosus, Ascophyllum nodosum, etc. Fucuccan sulfate participates in the occurrence of many diseases in the body and has various activities such as regulating blood sugar, anticoagulation, and anti-tumor. Based on this, it is very useful to develop sulfated fucan-based mimetics and apply it to drug and disease research.

Sulfated Fucan-based Mimetic Development Service at CD BioGlyco

  • Chemical Synthesis
The binding selectivity of polysaccharides mainly depends on their structural patterns such as sulfation pattern and alginate bond. Therefore, obtaining sulfated fucan-based mimetics with clear structure is crucial to studying the structure and activity relationship. Our synthesis group has extensive synthetic experience in providing precise and high-yield sulfated fucan-based mimetic synthesis services.
    • Protective deprotection chemistry and post-sulfation strategy: synthesis by free radical polymerization (CMRP) mediated by cyano group.
    • Microwave-assisted emulsion system: In a stable emulsion system, we provide facile and efficient synthesis service by microwave-assisted unprotected group open-ring reduction polymerization (ROMP).
  • Activity Analysis
Based on the synthetic sulfated fucan-based mimetics for different models, we provide corresponding analytical detection services.
    • In vitro inhibition of influenza A virus (IAV)
    • Inhibiting HSV-1 binding and entry into cells
    • Chronic renal failure (CRF)
    • Platelet cellular and molecular responses
    • Human breast cancer MCF-7 cells
    • Kidney disease model

Results: In this study, some fucoidan-mimetic glycopolymers are synthesized by RAFT polymerization. These copolymers have a homogeneous carbohydrate composition with different chain lengths. Researchers established a new method for detecting molecular markers for platelet aggregation and sections based on flow cytometry, and the samples were analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and western blotting. Studies have shown that tirofiban eliminates platelet aggregation stimulated by glycopolymers. In summary, the results show that synthetic fucoidan mimic glycocopolymers induce intracellular signaling, and significant changes in the biological surface, including conformational changes in αIIbβIII, leading to platelet aggregation and secretion of dense granules.

Applications of Sulfated Fucan-based Mimetic

  • Sulfated fucan-based mimetics is a unique tool to plays an important role in elucidating structure-activity relationships, cell surface interactions, and intracellular responses.
  • Sulfated fucan-based mimetics is effective in vitro and in vivo studies in the treatment of chronic renal failure, acute kidney injury, and diabetic nephropathy.
  • The sulfated fucan-based mimetic with different sulfation patterns is highly correlated with the antitumor activity of human breast cancer MCF-7 cells.
  • Sulfated fucan-based mimetics affect IAV infection by targeting the viral neuraminidase and the cellular epidermal growth factor receptor pathways. It provides a rational basis for antiviral drug development based on sulfated fucan-based mimics synthesized from marine sources.
  • A sulfated fucan-based mimetic is an excellent tool for establishing central pharmacological characteristics of receptor affinity, potency, and efficacy.
