Extraction and Purification of Chitosan

This process consists of demineralization, deproteination, and deacetylation. We extract chitosan from a variety of crustaceans and prepare it chemically, enzymatically, with ultrasound-assisted extraction, etc. In addition to this, a decolorization process is performed to remove the pigments contained in the samples. Differences in the chitosan preparation process and many parameters in the deacetylation reaction affect the properties of the final chitosan. Characterization of Chitosan

We characterize the physicochemical and functional properties of chitosan, including the degree of acetylation, distribution of acetyl groups along the chain, molecular weight, viscosity in solution, etc. Characterization protocols are customized to specific needs and include the following components:

  • Molecular weight: We examine molecular weight distribution, average molecular weight, and intrinsic viscosity.
  • Degree of acetylation (DA): DA is the proportion of acetylglucosamine units in chitosan and is influenced by various parameters in the deacetylation step. It affects a variety of physicochemical and biological properties of chitosan, such as hydrophilicity, crystallinity, and solubility. Infrared spectroscopy (IR), nuclear magnetic resonance (NMR), etc., are used to analyze the degree of acetylation of chitosan.
  • Acetyl group distribution: The distribution of acetyl groups along the chitosan chain is determined by 13C-NMR spectroscopy.
  • Crystal structure: X-ray diffraction, NMR, and IR are used to analyze the crystal structure of chitosan.
  • Functional groups: Fourier transform infrared spectroscopy (FTIR) is used to determine and analyze the functional groups in chitosan.
  • Scanning electron microscope (SEM): We analyze the surface morphology of chitosan by SEM to reveal the details of its microstructure.
  • Thermal stability: We analyze the thermal stability of chitosan by differential scanning calorimetry (DSC) and thermogravimetric analysis (TGA).

Chemical Modification of Chitosan

Results: In this study, a new method for the preparation of chitosan from shrimp shells was developed and the structure of chitosan was characterized by viscosimetry, XRD, UV, FTIR, and SEM. The results showed that the extracted chitosan samples were properly characterized, with their acetylation degree below 9%, ash content all below 0.063%, and soluble in 1% acetic acid. The molecular weights of the chitosan samples were found to be in the range of 2.3 and 2.8 × 105 g/mol and the degree of crystallinity was found to be around 40%. The results demonstrate that the extraction method developed in this study is suitable for obtaining chitosan with properties suitable for drug research. The method used in this paper also guides the development of extraction methods and structural characterization of chitosan.
