Process of Analysis
Analyzing the structure of marine glycoproteins is an important prerequisite for understanding their biological functions. We perfect the process of analyzing the structure of glycoproteins. We first use protease to cleave specifically modified marine glycoproteins into small fragments of glycopeptides, and then purify and enrich the glycopeptides obtained. Afterward, liquid chromatography-tandem mass spectrometry (LC-MS/MS) or hydrophilic interaction chromatography-tandem mass spectrometry (HILIC-MS/MS) is utilized for online sampling to obtain raw mass spectral data. Then, the obtained glycopeptide sequences are searched and analyzed based on compositional and structural fingerprints, and site and structure specificity databases. Finally, the structural analysis of marine glycoproteins is realized through the analysis of monosaccharides, molecular weight, amino acid composition, glycopeptide bonds, etc.
We have advanced LC-MS/MS, HILIC-MS/MS, and other high-resolution mass spectrometry systems. These advanced mass spectrometry systems support us in providing highly accurate glycoprotein structure analysis services.

Results: In this study, a glycoprotein (SGP1) with a molecular weight of 9.26 kDa was isolated from Sipunculus nudus. The structural analysis of this glycoprotein showed that the sugar moiety of SGP1 was mainly composed of glucose, xylose, and lysosaccharides. FT-IR results showed that SGP1 had typical glycoprotein features. It contains O-type glycosidic bonds and is a protein-dominated glycoprotein (92.8% protein).
