Marine Collagen Analysis Service at CD BioGlyco
CD BioGlyco provides clients with high-quality Marine Glycoprotein Analysis Services including physical, biochemical, and impurity analysis. Collagen is a trimeric molecule that has a variety of biomedical applications including bone and tissue regeneration, drug delivery, and wound healing. The properties of extracted collagen determine its quality and application potential. In addition to Quantitative Analysis and Structural Analysis Services for Marine Glycoproteins, we also offer analytical services for collagen derived from jellyfish, sponges, fish, and other invertebrates. We characterize collagen for various properties such as solubility, amino acid composition, morphological properties, structural properties, percent yield, molecular weight, viscosity, etc.
- Chemical composition of collagen: We use Fourier transform infrared spectroscopy to assess and identify the presence, chemical composition, and type of collagen. In addition to this, we use this method to study the effect of separation methods on collagen composition or to compare the composition of collagen extracted by different methods.
- Characterization purity and decomposition of collagen: We use sodium dodecyl sulfate-polyacrylamide gel electrophoresis to characterize collagen patterns and determine the molecular weight distribution of collagen peptides. We use this tool to separate proteins and their fragments from each other based on their size. When collagen bands are similar, we identify collagen types by comparing collagen obtained from other sources as a reference.
- Secondary structure of collagen: We evaluate the secondary structure, binding, and folding characteristics of collagen by circular dichroism.
- Morphological structure of collagen: We use scanning electron microscopy (SEM) to assess the morphological properties of collagen. The common form of collagen used for SEM analysis is powdered collagen obtained after freeze-drying.
- Collagen amino acid analysis: We use chromatographic techniques to separate, identify, and quantify amino acids. We will differences in the amino acid composition and content of collagen in different samples.
- Thermal characterization of collagen: High temperatures cause the molecular structure of collagen to unfold. We use differential scanning calorimetry to evaluate the thermal properties of collagen. The maximum transition temperature and denaturation temperature are also measured.
- Other properties of collagen: We also test collagen's viscosity, water-holding capacity, solubility, water absorption, etc.

Results: In this study, two collagens, including pepsin-soluble collagen (PSC) and acid-soluble collagen (ASC), were extracted from Nibea japonica swim bladders. The physicochemical properties of these two collagens, including amino acid composition, molecular weight, and others were analyzed. The results showed that both PSC and ASC contained β-chains, α-chains, and a small amount of γ-chains. Glycine accounted for nearly one-third of the total amino acid content of PSC and ASC. Both collagens have very similar thermal transition curves and the same surface morphology. They are suitable for drug delivery or wound healing.
