Pretreatment

  • Raw material selection: We carefully select algae varieties with rich lectin content and excellent biological activity as raw materials.
  • Washing and crushing: We thoroughly wash the selected algae raw materials to remove the mud and impurities on the surface. Subsequently, we crush the algae cells using physical or chemical methods to release the lectin in the cells.

Extraction

  • Solvent selection: We select the appropriate solvent for extraction based on the solubility and stability of the lectin. Commonly used solvents include water, salt solution, or organic solvents.
  • Extraction method: We use advanced extraction techniques such as ultrasound-assisted extraction, microwave-assisted extraction, or supercritical fluid extraction for extraction. During the extraction process, we strictly control parameters such as temperature, pH value, extraction time, and solvent concentration to maximize the retention of lectin biological activity and structural integrity.

Purification

  • Preliminary filtration: We remove large particle impurities in the extract by centrifugation and filtration.
  • Chromatographic separation: We use chromatographic columns to separate the extract according to the molecular weight, charge, or polarity differences between lectin and other components.
  • Gel filtration: We use gel filtration chromatography to separate lectin from other components according to molecular weight.
  • Ion exchange: We use ion exchange resin or membrane separation technology to remove ionic impurities in the extract.

Structural and functional characterization

  • Spectroscopic analysis: We use infrared spectroscopy, ultraviolet spectroscopy, and other technologies to analyze the molecular structure, chemical bonds, and functional groups of lectin.
  • Mass spectrometry (MS): We use MS to determine the molecular weight, amino acid sequence, and other information about lectin.
  • X-ray diffraction: We use X-ray diffraction to analyze the crystal structure and spatial arrangement of lectin.
  • Microscopic observation: We use electron microscopy to observe the morphology and microstructure of lectin.
  • Specific binding ability: We determine the binding ability and affinity constant of lectin for specific sugar molecules.
  • Signal transduction research: We study the potential mechanism of action of lectin in cell signal transduction.

Results: The authors discussed both traditional and more recent methods for extracting algal lectins, as well as the process of large-scale production. They also considered the difficulties and potential benefits of different extraction techniques. It was found that marine algae are a valuable source of unique lectins with various beneficial properties such as antiviral, antitumor, and antibacterial effects. One method discussed by the authors involved non-destructive extraction using algal culture water, which could allow for a consistent supply of lectins without harming the marine algae.
