  • Material pre-processing: We extract intact glycoproteins from different marine sources (plants and animals), including cell lysis, extraction, purification, and concentration of glycoproteins.
  • Release of glycans: This process involves the release of glycans from glycoproteins either chemically or enzymatically.
  • Derivatization of glycans: We perform fluorescent labeling or other derivatization of glycans to enhance the mass spectrometric response of glycans for identification.
  • Glycan analysis: Glycans are analyzed by liquid chromatography-mass spectrometry (LC-MS) and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS). Afterward, the relative contents of different glycan types are quantified according to the corresponding mass spectrometry peak areas, and bioinformatics tools are used to identify the glycan types and obtain the fine structure information of glycan chains.

Results: This study analyzes in detail the N-glycans and N-glycoproteins in Phaeodactylum tricornutum. The researchers identified 59 glycoproteins corresponding to N-glycans (mixed and high-mannose type N-glycans) through a series of treatments of extraction, digestion, enrichment, and labeling. Among them, the high-mannose-type N-glycans contained four types and all carried terminal glucose residues. This study provides a demonstration of the method for the analysis of N-glycan structures and updates the database of N-glycans in microalgae. It also provides a theoretical basis for elucidating the functions of these N-glycoproteins and for designing the use of microalgae for N-glycoprotein production.
