Raw material preparation and pretreatment

  • Raw material selection: We select high-quality S. filiformis with a high concentration of lectin as the raw material for extracting lectin.
  • Raw material pretreatment: Before the raw material is extracted, we perform appropriate pretreatment, such as washing, drying, and crushing to optimize the subsequent extraction conditions.

Extraction

  • Solvent extraction: We use a specific solvent system to effectively dissolve and extract lectin from S. filiformis by controlling factors such as temperature, pH value, and extraction time.
  • Optimizing extraction conditions: We conduct many experiments to determine the best extraction conditions, including solvent concentration, extraction temperature, and time to maximize the extraction rate of lectin.

Purification

  • Preliminary filtration: The extract is first filtered to remove large particle impurities and undissolved substances.
  • Chromatographic technology: We use advanced chromatography technology, such as gel filtration chromatography or ion exchange chromatography to further purify the extract to isolate high-purity lectin.
  • Ultrafiltration and concentration: We use ultrafiltration technology to remove small molecule impurities and concentrate the lectin solution to increase the concentration of the product.

Quality inspection and control

  • Activity detection: We use specific bioactivity detection methods to ensure that the extracted and purified lectin maintains its biological activity.
  • Purity analysis: Through high-performance liquid chromatography (HPLC), mass spectrometry (MS), and other technical means, the purity of lectin is analyzed in detail to ensure that the product meets high standards.
  • Safety assessment: We conduct strict safety assessments on the final product, such as microbial contamination detection and endotoxin detection to ensure the safety of the product.

Technology: Structural and activity characterization of S. filiformis lectin

Published: 2018

Results: The authors examined two variants of S. filiformis lectin (SfL), known as SfL-1 and SfL-2. The primary structures of these variants consisted of four tandem-repeat protein domains, each containing 67 amino acids. Analysis of the 3D structure predicted that both SfL-1 and SfL-2 were comprised of two β-barrel-like domains created from five antiparallel β-strands. Testing revealed that the mixture of isoforms (SfLs) exhibited an anticancer effect on MCF-7 cells.
