Keratan Sulfate Characterization Service

Keratan Sulfate Characterization Service

Unveil the Invisible- Keratan Sulfate (KS)

CD BioGlyco provides preparation, characterization, and analytical services for Polysaccharides from Different Marine Animal sources. KS does not contain glucuronic acid and consists of repeating disaccharide units that are involved in several physiological processes in the body. Combining a variety of advanced techniques and analytical tools, we offer a customized KS characterization service and provide detailed analytical reports. We not only help to analyze the conformational relationship of KS but also support the study of its role in various biological processes and diseases.

We isolate and purify KS from the tissues of marine animals according to the needs of our clients and determine its structural characteristics by various means of structural and compositional analysis. In addition to this, we also provide KS functional activity research services. Specific services include the following:

Extraction

KS is present in tissues such as the brain, cartilage, and cornea of marine organisms, and the degree of sulfation varies. Based on our extensive experience and efficient technology, we apply various strategies to the isolation of KS and optimize experiments to improve yield and purity. We design the best solution for KS preparation to meet your research needs.

Purification

We further purify KS by column chromatography, concentration, desalting, and other steps according to the properties of KS, and detect the products by thin-layer chromatography. Homogeneous polysaccharides are obtained by removing impurities for subsequent characterization and bioactivity analysis.

Characterization

We combine a variety of tools to analyze the structure, composition, and properties of KS, including nuclear magnetic resonance (NMR), liquid chromatography-mass spectrometry (LC-MS), infrared spectroscopy(IR), etc. We have extensive practical experience in structural and characterization analysis and have established a range of inspection methods and complete inspection processes.

  • Molecular weight and chemical composition analysis: We test for glucuronic acid and sulfate content. The purity and relative molecular weight of KS are determined by the high-performance gel filtration chromatography system. Monosaccharide composition is determined by high-performance liquid chromatography (HPLC).
  • Structural analysis: We analyze the type of glycosidic bond, functional group, degree of sulfated substitution, and others by NMR, IR, etc. Also, surface structure, micro-morphology, chain conformation, etc., are analyzed.

Results: In this study, a KS was extracted and purified from blue shark cartilage, and KS oligosaccharides were prepared by enzyme-catalyzed hydrolysis. The properties and structure of these two polysaccharides were also characterized. The results showed that the KS was highly sulfated and its non-reducing end was capped by N-acetylneuraminic acid. It had a binding affinity for Ricinus communis agglutinin I in a concentration-dependent manner. These results provide new structural features for the study of KS from blue shark cartilage and data support for the study of its potential application in ricin antidote development, as well as guidance for the preparation, characterization, and analysis of KS.

Frequently Asked Questions

What types of samples is the KS characterization service suitable for?

We analyze purified KS samples or KS-containing bio-biological matrices from various marine sources (e.g. corneal tissue, cartilage). Clients simply tell us their experimental goals and letter information. Our researchers will develop a customized protocol based on the type of sample and research objectives of the client, and complete the extraction, purification, and characterization experiments.

How long does the KS characterization service take?

The time required for KS characterization depends on the type of sample, the complexity of the analysis, and the techniques used. After understanding the client's needs we will design a detailed plan, provide an estimated project turnaround time, and efficiently complete the experiments to expedite the client's research.
