An aqueous extract of Cinachyrella is obtained by making a homogenate of the Cinachyrella sample with water and then lyophilizing it. The aqueous extract is then filtered through a cellulose membrane and dialyzed using water, and the substance obtained after dialysis is lyophilized to obtain crude Cinachyrella galectins.
Thecrude Cinachyrella galectins are initially purified on an ion-exchange column, and then the galectin fraction is further purified using an affinity chromatography column. In this process, tris-buffered saline (TBS) buffer is used to elute the proteins bound to the chromatography column from the galectin crude.
The product purified by affinity chromatography is subsequently purified in reversed-phase high-performance liquid chromatography (RP-HPLC), an operation that efficiently separates lactose-affinity proteins. Results: In this article, the authors describe the purification and analysis of Cinachyrella galectins. The galectins were efficiently purified from Cinachyrella using affinity chromatography, and X-ray crystallographic analysis revealed that Cinachyrella galectins possess a stable tetrameric structure formed by two sequences. In addition, Cinachyrella galectins were found to be active at high temperatures.
