Cinachyrella Galectin Production Service

Cinachyrella Galectin Production Service

Unleashing the Power of Nature: Premium Cinachyrella Galectin Production

Galectins are a kind of lectin that can recognize glycans containing β-galactose structure and is widely distributed in different kinds of organisms. Galectins have various and rich physiological activities, especially in the immune system. The special living environment and physiological structure of marine organisms make them an important source of lectins. Among them, Lectins from Sponges have favorable biological activity, such as anti-inflammatory and promoting cell recognition.

CD BioGlyco has in-depth experience in the field of Lectin Production and offers high-quality Cinachyrella galectin production service to clients with state-of-the-art lectin production and characterization technologies.

Extraction of Cinachyrella galectins

An aqueous extract of Cinachyrella is obtained by making a homogenate of the Cinachyrella sample with water and then lyophilizing it. The aqueous extract is then filtered through a cellulose membrane and dialyzed using water, and the substance obtained after dialysis is lyophilized to obtain crude Cinachyrella galectins.

Isolation and purification of Cinachyrella galectins

Thecrude Cinachyrella galectins are initially purified on an ion-exchange column, and then the galectin fraction is further purified using an affinity chromatography column. In this process, tris-buffered saline (TBS) buffer is used to elute the proteins bound to the chromatography column from the galectin crude.

The product purified by affinity chromatography is subsequently purified in reversed-phase high-performance liquid chromatography (RP-HPLC), an operation that efficiently separates lactose-affinity proteins. Finally, the galectin fraction is collected and the solvent is removed to obtain the pure galectin product.

Throughout the process, we use sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) to monitor the purity of galectins.

Analysis of Cinachyrella galectins

  • Structural characterization

We offer infrared (IR), nuclear magnetic resonance (NMR), and mass spectrometry (MS) to examine the primary and secondary structure of Cinachyrella galectins. In addition, the molecular ionic structure of Cinachyrella galectins can be accurately measured using, matrix-assisted laser desorption ionization- time-of-flight MS (MALDI-TOF-MS).

  • Molecular weight detection
    • Size exclusion chromatography offers a rough estimate of the molecular weight size of Cinachyrella galectins.
    • SDS-PAGE (using Caumas Brilliant Blue staining) and MS are two methods for efficiently measuring the molecular weight of Cinachyrella galectins.
  • Haemagglutination activity assay

Cinachyrella galectin is diluted with a specific buffer and tested for its haemagglutination activity in well plates. We add diluted Cinachyrella galectin, and erythrocyte suspension sequentially to the well plate and incubate it for a while to observe its haemagglutination activity under the microscope.

In addition, in the haemagglutination assay, we test the haemagglutination inhibition of Cinachyrella galectin by adding different kinds of glycans to it.

  • Cinachyrella galectin stability assay
    • The pH stability of Cinachyrella galectins can be tested by diluting Cinachyrella galectins with different pH buffers and performing a haemagglutination assay.
    • The temperature stability of Cinachyrella galectins can be tested by performing a haemagglutination test at different temperatures.

Results: In this article, the authors describe the purification and analysis of Cinachyrella galectins. The galectins were efficiently purified from Cinachyrella using affinity chromatography, and X-ray crystallographic analysis revealed that Cinachyrella galectins possess a stable tetrameric structure formed by two sequences. In addition, Cinachyrella galectins were found to be active at high temperatures.

Frequently Asked Questions

What types of cells can express galectins?

Galectins are expressed by activated T and B cells, regulatory T cells, dendritic cells, mast cells, eosinophils, monocytes/macrophages, and neutrophils because one of the galectins' main functions is to regulate the immune and inflammatory response.

What are the characteristics of galectins?

Galectins are found only in the animal kingdom and have preferential binding selectivity for galectin residues. Based on structural variability, galectins are classified into 3 subfamilies:

  • Prototypic galectins: They are non-covalent dimers, each with two identical carbohydrate recognition structural domains.
  • Chimeric galectins: They have two distinct structural domains that may bind the non-glycan portion to the glycan portion.
  • Tandem repeat galectins: They have two carbohydrate-recognizing structural domains linked by a functional linker peptide.
