Results: In this study, the researchers used ion exchange chromatography and sodium chloride gradient separation to obtain a HS-containing GAG extract from Portunus pelagicus. Fraction 5 (F5) was identified as HS by electrophoresis, FTIR, and NMR. The researchers tested the activity of F5 and found it to have potent beta-site amyloid precursor protein cleaving enzyme 1 (BACE1) inhibitory activity with highly attenuated anticoagulant activity. The mechanism of inhibition of the F5 is complex and different from that of heparin. This provides a potential starting point for marine GAG drug development.

What are the structural compositional peculiarities of HS?

HS is dominated by disaccharide units consisting of β-D-glucuronic acid and 2-N-acetylamino-2-deoxyα-D-glucose (including the O-sulfate substitution of C6) linked by (1→4). HS also contains β-D-glucuronic acid or aldose linked to 2-N-sulfo-2-deoxyα-D-glucose (including the O-sulfate substitution of C6).

What is the principle of polysaccharide separation on the DEAE-cellulose column?

DEAE-cellulose has an open backbone, and polysaccharides enter the carrier freely and diffuse rapidly. Its separation mechanism is not a single ion exchange, but adsorption and desorption. The components to be separated in the solution are adsorbed on the ion exchanger by the Coulomb force and charge difference, and the suitable eluent is selected to elute the adsorbent on the ion exchanger to achieve the purpose of separation. Using different ionic strength solutions to elute step by step, to get different components of polysaccharide and achieve the separation effect.
