Extraction and Isolation of Roselipin

  • Structural Analysis
    We use multiple types of nuclear magnetic resonance (NMR), infrared (IR) as well as ultraviolet spectroscopy (UV) to characterize the general structure of roselipin.
    Among them, heteronuclear multiple quantum coherence (HMQC) spectra analyze the connection between protons and carbon, and 1H-detected heteronuclear multiple bond correlation (HMBC) spectra present information about the coupling of carbon and hydrogen atoms.
    On the other hand, IR and UV spectra show us information about the specific groups contained in roselipin, including the type of group, the position of the linkage, and the conformation of the sugar ring, among others.
  • Molecular Weight (Mw) Analysis
    The relative Mw of roselipin is analyzed by gel permeation chromatography (GPC) and mass spectrometry (MS).
  • Purity Analysis
    The purity of roselipin is analysed by HPLC, i.e. the purity of roselipin is calculated from the area of the peaks in the spectrum.
  • Molecular Formula Analysis
    We provide the fast-atom-bombardment MS (FAB-MS) method to detect the molecular formula of the extract, which can further determine the accuracy of roselipin products.

Results: In this article, the authors show the origin and biological activity of roselipin, which was isolated from the fungus Gliocladium roseum KF-1040 and tested by various NMR and found to have a unique structure in which a highly methylated fatty acid backbone is modified by mannose and arabinitol residues. Biological evaluation of roselipin revealed superior inhibitory activity against diacylglycerol acyltransferase. In addition, roselipin showed some antibacterial activity against Saccharomyces cervisiae and Aspergillus niger.
