Extraction of Simplexide

We use methanol and trichloromethane as extraction solutions to extract simplexide sequentially from marine sponges. After the extraction is complete, all the extracts are combined and partitioned between water and n-BuOH. Then, the organic phase is collected and the solvent is removed under vacuum to obtain the crude extract of simplexide.

Purification of Simplexide

Simplexide is purified sequentially using reversed-phase column chromatography and direct column chromatography. Throughout the purification process, a thin-layer chromatographic (TLC) is used to check whether impurities are effectively removed. The purity of the obtained simplexide is tested by nuclear magnetic resonance hydrogen spectroscopy (1H NMR).
