Extraction of Ganglioside GP-3

  • GP-3 is extracted using a solvent mixture of methanol and chloroform in a certain proportion after chopping the whole body of the sea cucumber. After some time, the starfish residue is removed and the methanol and chloroform are removed in a vacuum to obtain a glycolipid-like mixture containing GP-3.
    The glycolipid-like mixture is extracted using AcOEt/n-BuOH to remove the less polar glycolipids. Where the aqueous layer is then sequentially extracted using a n-BuOH and a mixture of methanol and chloroform and all solvents are removed in vacuo to give a crude GP-3.
  • For the purification of GP-3, we perform a reversed-phase column chromatography. We use silica gel as the stationary phase of column chromatography, and different ratios of mixed solvents of methanol and chloroform as the eluent to purify GP-3. Meanwhile, the purified GP-3 showed a single spot on thin-layer chromatography (TLC).

Total Synthesis of Ganglioside GP-3

  • In general, the synthesis of ganglioside consists of two main parts: the construction of the glycan structure and the selection of the ceramide structure.
    Here, we perform the total synthesis of GP-3 by stereoselective and efficient glycosylation reactions. In this case, the construction of the glycan structure part is carried out by polymerization coupling operation. The glycan structure is used as a donor and conjugated with glucosylceramide acceptor to synthesize GP-3 in high yield.
  • The GP-3 is purified by reversed-phase silica gel column chromatography, in which a mixture of methanol and chloroform in different ratios is selected as the eluent. The purified GP-3 is a white solid.
