Extraction and Isolation of Vesparioside
We use a mixture of chloroform and methanol as an extraction solvent to extract vesparioside from the sponge Spheciospongia vesparia. At the end of the extraction, chloroform and methanol are removed to obtain the crude product of vesparioside.
Purification of Vesparioside
- The crude extract of vesparioside is extracted using a mixture of water and BuOH and the organic phase is collected.
- The organic phase is concentrated in a vacuum and then purified sequentially using silica gel and silica columns, a step that effectively removes non-glycolipid impurities.
- To keep the structure of vesparioside from being destroyed in the subsequent purification step, we perform acetylation of the glycolipid mixture. Afterward, high-performance liquid chromatography (HPLC) is used to remove other glycolipid compounds, which in combination with the deacetylation operation results in pure vesparioside.
Characterization of Vesparioside
- Structure
Vesparioside has a complex and specific structure, so we analyze its structure using microscale chemical degradation combined with mass spectrometry (MS), nuclear magnetic resonance (NMR), and circular dichroic (CD) spectrum. That is, we perform acidic decomposition of vesparioside and use MS, NMR, and CD spectrum to analyze the structure of the product and the conformation of the sugar chain. Finally, the structural characterization of vesparioside is deduced from all the results.
- Purity
We utilize HPLC to test the purity of vesparioside to ensure that the vesparioside produced meets the purity requirements of our clients.

Results: In this article, the authors show the structural features of the marine glycosphingolipid vesparioside, which was isolated from the marine sponge Spheciospongia vesparia after an organic solvent extraction method. After analysis using spectroscopic techniques, it was found to be a unique disaccharide glycosylated marine glycosphingolipid with fragments of furanose residues in its structure.
