Gangliosides are an important class of glycolipid molecules in the nervous system, and they are involved in the onset and progression of a variety of neurological diseases. Extraction and Purification of SJG-1

  • After cutting the body wall of the sea cucumber, SJG-1 is extracted from it using a mixture of CHCl3 and MeOH. After repeated extractions, all the extracts are combined and concentrated in a vacuum. Next, appropriate amounts of hexane, methanol, and acetone are added to it sequentially. The insoluble material is collected and dried to obtain the crude product of SJG-1 after stirring.
  • We utilize reversed-phase column chromatography to purify SJG-1. In this process, silica gel is used as the stationary phase, and a solvent mixture of MeOH, CHCl3, and H2O is used as the mobile phase to purify SJG-1.

Extraction and Purification of SJG-2

  • The extraction process of SJG-2 is similar to that of SJG-1. A certain ratio of trichloromethane and methanol is still used to extract from the body wall fragments of sea cucumbers.
  • For the purification of SJG-2, we use reverse-phase column chromatography, ion exchange chromatography (IEC), and gel filtration chromatography (GFC).
    First, the crude SJG-2 is placed in a silica gel column and eluted with methanol as eluent. After collecting the SJG-2-containing fraction, it is purified by an IEC and a GFC turn. Finally, the SJG-2 fraction is collected and dried in a vacuum to obtain pure SJG-2. In this process, we use thin-layer chromatography (TLC) to detect the purity of SJG-2, i.e., pure SJG-2 should have a spot on the TLC plate.

Characterization of SJG-1 and SJG-2

Both SJG-1 and SJG-2 have the appearance of an amorphous powder, and we offer characterization that includes, but is not limited to, the following:

  • Structure: Nuclear magnetic resonance (NMR) and infrared (IR) spectroscopy are powerful methods for characterizing SJG-1 and SJG-2. Among them, IR can effectively characterize the hydroxyl group and amido group of SJG-1 and SJG-2.
  • Relative molecular weight: The relative molecular weights of SJG-1 and SJG-2 are analyzed by fast-atom-bombardment mass spectrometry (FAMS).
  • Purity: The purity of SJG-1 and SJG-2 is determined using high-performance liquid chromatography (HPLC) and gas chromatography-mass spectrometry (GC-MS).

Results: In this article, the authors describe the origin and bioactivity of the ganglioside SJG. SJG exists in two structures, SJG-1 and SJG-2, and both were isolated from the marine organism sea cucumber using an organic solvent. After bioactivity testing, it was found that SJG-1 and SJG-2 showed neurogenic activity against rat pheochromocytoma PC12 cells. Moreover, the activity of SJG-2 was higher than that of mammalian gangliosides.

What is the structural difference between SJG-1 and SJG-2?

Both SJG-1 and SJG-2 are overall composed of two parts, the glycan structure and the ceramide structure. Compared to SJG-1, SJG-2 has a more complex glycan structure, i.e., SJG-2 has a special hexasaccharide chain: NeuAc2→3(NeuAc2→4)→Gal1→8NeuAc2→3GalNAc1→3Gal1→4Glc.
